Disentangling the mutational effects on protein stability and interaction of human MLH1

In this study, the authors use high-throughput experimental approaches to test how missense mutations affect the cellular abundance of MLH1 and the interaction with its partner, PMS2. They find that many variants disrupting the interaction with PMS2 also lead to reduced cellular abundance levels, suggesting a strong relationship between protein stability and interaction in MLH1.

Here, the authors focus on functional classes such as mammary-specific genes, lactation-associated genes, non-coding RNAs, RNA editing sites, DNA methylation and expression quantitative trait loci. Analyzing 3 million variants in 23,566 Holstein bulls, they identify key variants and functional classes that contribute significantly to genetic variation in milk production.

Exploring the molecular basis of the genetic correlation between body mass index and brain morphological traits

Here, the authors investigate the genetic variants that affect both obesity and brain morphology, identifying 21 genes whose expression affects both body mass index and morphological measures obtained from brain magnetic resonance imaging data. These results provide new insight into the complex relationships between the brain and obesity.

Genetic risk for neurodegenerative conditions is linked to disease-specific microglial pathways

In this study, the authors use a 3D genomic approach to map how genetic variants linked to neurodegenerative diseases interact with genes in different brain cell types. Their analysis reveals that genetic risk for Alzheimer’s disease, Parkinson’s disease, multiple sclerosis, and amyotrophic lateral sclerosis is particularly enriched in microglia, the brain’s immune cells.

Conserved chromatin regulators control the transcriptional immune response to intracellular pathogens in Caenorhabditis elegans

In this study, the authors show that transcriptional regulators, previously linked to development in Caenorhabditis elegans, also control immune responses. They also discovered that other chromatin regulators similarly repress IPR gene expression, highlighting a new role in immunity for these conserved regulators of development.

Germline mutation rates and fine-scale recombination parameters in zebra finch

Here, the authors characterize properties of mutation and recombination in the zebra finch, which, like other birds, differs from mammals in several potentially salient respects, including its karyotype and the mechanism by which recombination is directed to the genome. By sequencing the genomes of three-generation pedigrees, the authors identify de novo mutations and recombination events.
